Worked seemlessly. Now I want to configure a new Email Account, but I cannot find the settings to achieve that. The instructions say that there should be a button next to owl icon under addons, but there is not. Useless for me right now!

Everything works again as expected. I just updated to the newest version of Thunderbird (78.2.1) before the bug arose, but I don't think that the update is the source of the bug.
The source of the problem came from changing the profiles folder location, which made the settings tab disappear and removing my exchange account.
The settings tab for the addon was visible again, after I uninstalled, restarted thunderbird (restarting was necessary) and reinstalled the addon. The settings tab then showed an error that it couldn't find the addon's settings folder, pointing to the old profile location. After uninstalling the addon, restarting tb and installing again, the account showed up again and the addon worked as expected.

It appears that you cannot find the Owl settings page.
Unfortunately, Thunderbird (and Firefox) keeps changing the UI and buttons how to access it, with almost every version. So, it's difficult to tell in a generic way, and I need to know your Thunderbird version.
Generally, the Owl settings are accessible in the same way as the settings of all other TB extensions. In Thunderbird 78, depending on which exact view you are on, you need to click on the "..." button for Owl (in the addons list) or the "Preferences" tab (in the addon details page).
And, of course, please make sure that you have the latest version of the addon installed (currently version 0.7.6).
To create your own collections, you must have a Mozilla Add-ons account.
